The purpose of this study was to examine the effect of chronic molecular hydrogen (H2) intake on liver and skeletal muscle glycogen utilization during exercise in rats. Male SD rats were randomly divided into four groups: control (C), H2 water (H2), control-exercise (C-Ex), H2 water-exercise (H2-Ex). Each group was given mineral water or H2 water ad libitum for 2 weeks. Then the exercise groups (C-Ex and H2-Ex) were performed treadmill running at 18 m/min (6° graded) for 60 min. Blood, skeletal muscle and liver samples were analysed for tissue glycogen contents and blood energy substrats (i.e., glucose, free fatty acid, lactate). Two-way ANOVA revealed that H2 intake decreased free fatty acid (p < 0.001) and increased lactate concentration (p = 0.004). In comparison of the change from the rest in C-Ex and H2-Ex groups, blood free fatty acid and liver glycogen content were significantly lower in H2-Ex group than in the C-Ex group (p = 0.001, p = 0.035, respectively). On the other hand, blood lactate concentration was significantly higher in the H2-Ex group than in the C-Ex group. These results suggest that chronic H2 water intake may increase liver glycogen utilization without changing muscle glycogen utilization during exercise.
